Orthology assertions are a critical get comparative genomics and are challenging component on a genomic annotation, and it has been a large part of Xenbase and NCBI curators work to make and improve these assertions for the Xenopus genomes.

We recently realized that 2 genes were mis-annotated in the Xenopus v10 genomes: orai1 and orai2.

Phylogenetic analysis, synteny and reciprocal BLAST confirm that these two genes are misidentified as each other: true human ORAI1 orthologs were misidentified as orai2, and the true orthologs of human ORAI2 were misidentified as orai1.

The simple synteny maps on Xenbase (screen shots attached), and the phylograms below (the genes in the correct clades, but with the wrong gene symbol) confirm the error in gene name assignment.

The renamed Xenopus orai1 genes, which are all on chromosome 1/1L/1S are now represented on XB-GENEPAGE-5781665:

Xtr.orai1 Gene ID: 100380109 ENSXETG00000004483

Xla.orai1.L Gene ID: 108716807

Xla.orai1.S Gene ID: 108716807

The renamed Xenopus orai2 genes, which fall on chromosome 2/2L/2S, are now represent on XB-GENEPAGE- 962825.

Xtr.orai2 Gene ID: 100125061 ENSXETG00000043670

Xla.orai2.L Gene ID: 108707681

Xla.orai2.S Gene ID: 403390
